5’-AMP-activated protein kinase (AMPK) is a master sensor of energy regulating metabolic homeostasis. Recently, several studies reported that activation of AMPK restrict infectious viruses such as SARS-CoV-2, zika virus, Kaposi’s sarcoma-associated herpesvirus, and coxsakievirus B3. However, the antiviral effect of AMPK activation against hepatitis B virus (HBV) infection is not elucidated. Here, we investigated the anti-HBV activity of pharmacological activator of AMPK. 5-aminoimidazole-4-carboxamide-1-β-D-ribofuranoside (acadesine), analog of AMP used for activation of AMPK. To determine the HBV inhibitory concentration of acadesine without cell toxicity, HBV antigen secretion and MTT assay were assessed in HepG2.2.15 and HepG2 cells. HBV life cycle including HBeAg/HBsAg secretion, HBV transcription and replication were analyzed in HepG2-NTCP cells with HBV infection. Acadesine inhibited the HBV antigen secretion, especially surface antigen, in HBV expressing cell lines and attenuated HBV DNA and RNA in HBV infection system. The molecular mechanism of anti-HBV activity of acadesine was the activation of the MAPK (Erk1/2), consequently down-regulated the expression of HNF4α binding to HBV enhancers. These results indicated that acadesine inhibited HBV replication at the transcriptional level through activation of MAPK-HNFs signal pathway. Finally, HBV infection decreased AMPK phosphorylation, suggesting that AMPK activation may play a role of cellular restriction factor in HBV-infected hepatocytes. This study identified the possibility of acadesine as potential therapeutic agent for chronic hepatitis B
