This study aims to develop effective therapeutic agents for Lassa virus by systematically expanding viral libraries, construction of comprehensive immunogenic Lassa-Gpc antigen and specific antibody resources, and establishment of cell-based antiviral screening platforms for the identification of novel therapeutic candidates. Additionally, through international collaborations, the project will strengthen its research foundation and contribute to global efforts to combat Lassa virus infections. (Year 1: Expansion of Target Virus Library and Resource Development) The primary goal was to expand the Lassa virus evaluation library and develop essential research resources. This involved acquiring additional recombinant antigen clones from the recently emerged Lineage IV and VII strains, which were isolated from major outbreak regions during 2019-2020. By incorporating these new strains, it aimed to broaden the scope of viruses being evaluated for potential therapeutic interventions. A key focus was on constructing a comprehensive antigen library centered on the Lassa virus glycoprotein complex (GPC), which includes GP1, GP2, and GPC domains. This library was designed to include both genotype-specific (types 4-5) and cross-reactive antigens, facilitating more robust immunological screening. Simultaneously, monoclonal antibodies were generated through mouse immunization using Lassa GPC antigens, including recombinant antigen and Lassa-GPC pseudovirus lineage 2-4). This was resulted in the secureing LAV-Abs library with 52 hybridoma-derived mAbs, 9 of mouse scFv, and 9 phage-display based human IgG Abs. Additionally, we focused on improving pseudovirus production systems by employing lentivirus-based pseudovirus vectors and showed enhanced the production efficiency by around X10 fold compared to traditional systems, thus enabling high-throughput screening of antiviral compounds. In secondly main research was to validate and refine a cell-based antiviral screening platform. Using Lassa GPC pseudoviruses, a neutralization assay was established, which enabled the evaluation of antiviral efficacy against a library of genotype-specific monoclonal antibodies. This process led to the selection of 8 promising antibody candidates for further research. Furthermore, the project optimized a rapid, large-scale screening platform within a Biosafety Level 2 (BSL-2) facility. This platform, utilizing Lassa GPC pseudovirus, was designed to efficiently test a broad range of antiviral compounds. Additionally, high-throughput screening (HTS) was employed to identify novel antiviral candidates, including the testing of FDA-approved nucleic acid-based therapeutics to identify potential treatments for Lassa virus.
