SARS-CoV-2 belongs to the coronavirus genus and is characterized by high transmissibility and the emergence of continuous mutations. Therefore, there is a possibility of new coronavirus-derived diseases emerging in the future, and preparation for the next coronavirus pandemic is necessary. One of these preparatory measures is to study currently existing coronaviruses using clinical specimens from COVID-19 patients and animal models susceptible to coronavirus infection. Our researchers explored the pathogenic mechanisms and immune factors associated with coronavirus infection using clinical specimens from severe and critical COVID-19 patients and mouse models susceptible to coronavirus infection. When metabolomic analysis was conducted on clinical specimens from COVID-19 patients, we observed a decrease in various carnitines, glutamine, and malic acid, as well as an increase in linoleic acid, correlating with disease severity. Moreover, we noted that as the ratio of glutamic acid to glutamine increased, there was a tendency for disease severity to worsen. Furthermore, our research team conducted transcriptomic and metabolomic analyses following MERS and SARS-CoV-2 infections using hiACE2-hDPP4 mice, a model susceptible to various coronaviruses. As a result, we observed a decrease in arginine levels across all experimental groups, consistent with the metabolomic analysis of COVID-19 patients. Additionally, analysis of gene expression patterns related to arginine metabolism revealed that among seven relevant genes, four genes (Fah, Nos3, Pink1, Por) showed decreased expression in all experimental groups 6 days post-infection, while two genes (Mcm3, Zfand6) increased compared to the control group. The remaining gene (Xdh) increased only in the MERS experimental group, differing from the SARS-CoV-2 group. Therefore, through transcriptomic and metabolomic analyses of COVID-19 patients and coronavirus-infected mouse models, we expect that metabolites related to arginine biosynthesis may serve as metabolic indicators reflecting disease progression in coronavirus infection and potentially act as therapeutic targets.
