ㅇ Development of a multivalent vaccine platform for controlling co-infection ▸Development of influenza virus-vectored multivalent vaccine platform - A novel multivalent vaccine platform has been developed using influenza virus vectors to control co-infections of SARS-CoV-2 and influenza. This platform incorporates nine NS segments with the SARS-CoV-2 RBD domain and has resulted in the generation of five recombinant vaccine candidates. These candidates were evaluated for growth kinetics in embryonated eggs, interferon sensitivity, and pathogenicity in animal models. Optimal immunization conditions, including vaccination routes, doses, and the use of immunoadjuvants, were established through an analysis of binding and neutralizing antibody responses. A single final vaccine candidate was selected based on these parameters. Vaccination with this candidate demonstrated reduced lung viral titers, and efficacy was further enhanced with an RBD protein booster. Additionally, genetic analysis through serial passaging confirmed the genetic stability of the influenza virus-vectored system. ▸Development of bacteria-like particle multivalent vaccine platform (collaboration w/ POSTECH) - A multivalent vaccine platform was also developed using bacterial-like particles. This approach involved conjugating plant-derived SARS-CoV-2 S and influenza HA proteins to inactivated bacterial BLPs, with each particle carrying a single antigen. The mixed BLPs were used to generate vaccine candidates. Immunogenicity analyses, including binding and neutralizing antibody titers, were conducted to optimize immunization parameters such as the route and dose of administration. BLPs proved to be effective adjuvants, providing robust immune responses. Under optimized conditions, the vaccine demonstrated 100% survival rates in both single and co-infection models, even with minimal doses of SARS-CoV-2 S(1 μg) and influenza HA(1 μg). ㅇ Investigation of host factors for developing optimal(susceptivble) animal models for co-infection - Innate immune factors that regulate co-infections of SARS-CoV-2 and influenza virus were identified and characterized. Using CRISPR knockout screening and single-cell RNA sequencing, researchers identified innate immune sensors involved in pyroptosis during SARS-CoV-2 infection. These sensors were further studied to elucidate their protective roles in vivo and to understand the inflammatory pathways they mediate. ※ This can serve as foundational data and technology for addressing co-infections in volving emerging respiratory viruses
