371주, Salmonella spp. Salmonella spp.
Collection and Resistance of Clinical Isolates of Campylobacter and Salmonella spp. - As a result of collecting clinical isolates derived from acute diarrhea patients who visited 106 primary and secondary medical institutions in 6 regions nationwide from April to September 2022, Campylobacter spp. 371 isolates and Salmonella spp. 437 isolates were collected. As the main antimicrobial resistance, 83.3% of resistance was confirmed in fluoroquinolone and 32.3% in tetracycline. Salmonella spp. 437 isolates showed overall multidrug resistance (MDR) in various antibiotics, especially in Colistin, 60 isolates out of 437 isolates, 13.7%, showed resistance. Even after the ban on ciprofloxacin use in 2010 in South Korea, no rapid decline could be observed and high-level ciprofloxacin resistance was still identified. Fluoroquinolone resistance is believed to be maintained due to the increased use of enrofloxacin in farms until 2022. Clonal population - Through MLST results, 57 STs and 20 CCs were identified in C. jejuni, and the main clonal types were CC21, CC22, and CC206, and C. coli mainly had CC828 clonal types. Salmonella spp. detected 55 STs in 437 isolates, and the main epidemic clones were identified as ST11, ST34, and ST32. This study highlights that quinolone- and tetracycline-resistant Campylobacter circulate in Korea with diverse genotypes. Characterization of patient-derived resistant genomes - Based on the results of antimicrobial resistance, 40 C. jejuni, 10 C. coli, 10 S. Enteritidis, and 1 S. Bareilly were selected to secure the human-derived resistant bacterial genome through full-length genome analysis, and the results were derived by analyzing major antibiotic resistance genes, virulence factors, and resistance cause characteristics. Analysis of the mechanism of Salmonella colistin resistance - In order to study the colistin resistance mechanisms of the two serotypes of S. Enteritidis and S. Bareilly, whole genome & transcriptome-based analysis revealed that both mcr genes were negative, but colistin resistance strain-specific mutations were identified in the ann operon and rfa genes, which are important genes related to LPS modification that play an important role in the mechanism of colistin resistance. In addition, various mutations were found in O-antigen-related genes (rfaL, wzzB) and MDR efflux, OMP, and TCS, respectively. - As a result of transcriptomic analysis, new resistance cause candidate genes related to colistin resistance mechanisms were derived as they showed high expression in related genes such as two-component systems (TCS) and cationic antibacterial peptide (CAMP).
