Research background & objective ㅇ The Nipah(NiV) causes annual outbreaks in Southeast Asia, resulting in a mortality rate of 40-70%. In the absence of licensed vaccines or specific therapeutics, NiV is listed as a priority pathogen of epidemic and pandemic potential by WHO(2023). Nanobodies(VHH), are unique class of antigen-binding fragments derived from camelids and cartilage fish antibodies, known for their strong binding affinity, high stability, and tissue penetration due to their small size(12~15kDa). Their structure enables easier genetic modification and protein engineering, compared to conventional antibodies. Synthetic nanobody libraries are increasingly used to identify high-affinity binders, with numerous neutralizing nanobodies developed against SARS-CoV-2. Here, this proposal aims to rapidly identify synthetic nanobodies targeting NiV, with the goal of selecting potent neutralizers. Results and Conclusion ㅇ First, the fusion F protein ectodomain encoded by a relevant NiV strain was used to design and produce a stabilized trimeric prefusion form of the protein (preF). In parallel, an infectious pseudotype NiV(NiVpp) was generated using either NiV-F + NiV-G surface antigens, or a Vesicular Stomatitis Virus G surface antigen (VSV-G) as VSVpp control. NiVpp was highly infectious, and the infectivity was specifically blocked by the 5B3 reference antibody. In contrast, purified preF was able to block the antibody-mediated neutralization, validating its use in nanobody screening. By using preF as a bait, a synthetic nanobody library was screened by ribosome-display. After 7 rounds of enrichment, a comparison of amplified nanobody sequence clusters led to the selection of 31 potential binders. Of these, 15 sequence-optimized nanobody clones were successfully expressed in the mammalian system and purified, and their NiVpp-neutralizing ability, along with preF bidning capacity, were assessed. As a result, nanobody Top19 exhibited NiVpp-neutralizing activity at 400μg/mL of concentration. However, similar neutralizing activity was also observed against VSVpp, indicating that a potential broad-spectrum activity that requires further investigation. Additionally, the size-exclusion chromatography profiles and SDS-PAGE analysis revealed that nanobody Top19 existed in 2 oligomeric states as well as 2 possible sugar binding forms. By using chemical protein crosslinking, Top19 and Top7 nanobodies were found to be in complexes with preF, but not Top2. Overall, the results illustrated that it is possible to produce a functional Nipah virus antigen and identify nanobodies able to bind (at least 2 clones) to preF, and to neutralize NiV pseudotyped virus (1 among 13 clones tested), in a relatively short period of time. Such a platform could be instrumental for epidemic and pandemic preparedness, not only for the development of therapeutics, but also of diagnostic tools.
