In the first year, we attempted to conduct prophage transfer targeting various antibiotics by antibiotic class, and E. coli was divided into five groups. Group A includes O157 and induces HUS. Group B is associated with HUS, but is less common than O157:H7. Group C is associated with sporadic HUS, group D causes diarrhea but is not associated with HUS, and group E does not cause human disease. However, in the second year, the principal investigator changed, and antibiotics were limited to empirical antibiotics mainly used for diarrhea patients, and E. coli was divided into three groups according to the frequency of occurrence worldwide to conduct prophage transfer research. Prophage Dorner and Recipient groups of E. coli measured MIC for five antibiotics, and based on this, the antibiotic concentration was determined in subsequent studies. The expression of toxin genes according to antibiotic concentration showed an increase at 1/4 MIC concentration in Ciprofloxacin and Levofloxacin, but in the case of Azithromycin, the expression of toxin genes did not increase. Similar to the results of toxin gene expression, high transfer was confirmed in Ciprofloxacin and Levofloxacin, and low transfer was confirmed in the case of Azithromycin. In addition, it was revealed that the serotype of the Recipient played a more important role in transfer than the serotype of the Dorner. In addition, it was proven that the chromosome insertion site of the transferred prophage was inserted into the four main sites after transfer. In a mouse infection experiment, Ciprofloxacin was confirmed to induce 57 times more metastasis than Azithromycin, and in this study, it was confirmed that it was transferred to 20 different serotypes. In addition, the transferred toxin gene showed various patterns of cytotoxicity, but further research is needed to determine the exact mechanism.
