Purpose : Rotaviruses are the major etiologic agents of acute gastroenteritis worldwide. The evaluation of performance of a rotavirus vaccine depends on the knowledge of the epidemiological features of rotavirus infection in a regional setting. The purpose of this study was to analyse prevalent genotypes in Korea and develop PCR-ELISA for identification of G and P genotypes of group A rotavirus. Methods : To investigate the genotypic distribution of rotavirus strains in Korea, we analyzed VP4 and VP7 regions of 1,170 samples(399, 280, 491 samples from 2005 to 2007, respectively). G and P typing was subsequently carried out by genotype specific nested PCR. We determined the VP4 and VP7 gene by automatic sequencing and analysed sequences using DNAstar software. Results : The results showed that 4 major genotypes: P[8]G1, P[8]G3, P[4]G2, and P[6]G4 were found to be prevalent in Korea, although relative frequencies of those 4 types changed during this period. The genotypic pattern of rotavirus is changing continuously in Korea. And we selected that G- and P- type specific primer and probe by comparison with VP4 and VP7 sequences of Korean strains. Conclusions : In the study, we analysed G- and P- genotypic distribution of group A rotavirus in Korea from 2005 to 2007 and selected that G- and P- type specific primer and probe using PCR-ELISA for identification of human group A rotavirus genotypes.
