전분화능줄기세포를 이용한 인간화 간(humanized liver) 마우스 개발

초록

ㅇ 연구개발 목표 전분화능줄기세포 유래 인간 간세포를 지닌 인간화 간 마우스 개발을 목표로, 면역결핍마우스에 전분화능줄기세포 유래 인간화 간세포 이식을 위한 최적화연구를 수행함으로써 HBV 및 HCV 감염 질병 모델을 개발함 ㅇ 연구개발 내용 -1차년도 ◦ 인간 간세포 이식용 FAH 결손 면역결핍마우스 생산 ◦ 간세포 이식을 위해 면역결핍마우스 간세포 사멸 조건 및 이식 조건 탐색 ◦ 인간 primary 간세포 (PHH) 확보 ◦ 간세포 이식 마우스에서 혈액 내 마우스/인간 알부민 분포도 측정 ◦ 전분화능줄기세포 유래 인간 간세포 분화기술 확보 - 2차년도 ◦ 세포 유래 및 이식 조건별 인간화 정도 분석을 통한 이식 조건 고도화 ◦ 전분화능줄기세포 유래 인간-간 유사세포 분화 위한 최적 조건 확보 ◦ 인간화 간 마우스에서 간조직의 물질대사 분석 ◦ HBV 감염 테스트를 위한 인간화 간 마우스 제공(5마리/회x3회 이상) - 3차년도 ◦ 사람 간세포(primary hepatocyte) 및 전분화능줄기세포를 분화시킨 간세포 이식 마우스 간조직의 인간화 비교(HBV/HCV 감염 마커 포함) ◦ 세포유래 및 이식조건별 인간화 정도 분석을 통한 이식 조건 최적화 ◦ 전분화능줄기세포 유래 인간화 간 마우스 제작 및 유지관리 최적화 ◦ 인간화 간 마우스의 간세포에 대한 scRNA-seq 등 NGS 분석 ◦ HCV 감염 테스트 ◦ HBV 감염 테스트/연구를 위한 인간화 간 마우스 제공 (5마리/회x3회 이상) ㅇ 연구개발 성과 - 인간화 간 마우스 상업화 - 인간화 간 마우스를 만성병 및 감염병 질병모델 연구에 활용 - 간질환관련 기초연구는 물론 신약개발을 위한 비임상모델동물로 활용

ㅇ Purpose The purpose of this research is to develop humanized liver mice using human hepatocytes derived from pluripotent stem cells and primary human hepatocytes that can be infected by HBV or HCV ㅇ Contents The 1st year ◦ Production of immune deficient mice deleted FAH for engraftment of human hepatocytes ◦ Investigation of conditions of hepatocyte cell death and transplantation in immune deficient mice for engraftment of human hepatocytes ◦ Securing primary human hepatocytes ◦ Measurement of human albumin level in blood of the mice engrafted human hepatocytes The 2nd year ◦ Optimization of conditions of transplantation through analysis of humanization levels according to conditions of origin and transplantation of cells ◦ Establishment of optimal conditions of hepatocyte differentiation from pluripotent stem cells ◦ Analysis of metabolites of liver tissues in humanized liver mice ◦ Provision of humanized liver mice for testing infection of HBV (5 mice every time, more than 3 times) The 3rd year ◦ Comparison of the level of humanization between the mice engrafted with human hepatocytes differentiated from pluripotent stem cells and the mice engrafted with primary human hepatocytes (including markers for infection of HBV/HCV) ◦ Optimization of humanized liver mouse production and management of humanized liver mice derived from pluripotent stem cells ◦ Analysis of NGS on hepatocytes of humanized liver mice ◦ Efficacy test of HCV infection with humanized liver mice ◦ Provision of humanized liver mice for testing infection of HBV (5 mice every time, more than 3 times) ㅇ Development Results ◦ Commercialization of humanized liver miceion ◦ Application for the research of infectious disease model and chronic disease using humanized liver mice ◦ Application with preclinical model animals for development new drugs as well as basic research associated with liver diseases
