Establishment of a Neutralization Potency Evaluation Platform for COVID-19 Antibodies ◈ Objective: To establish a rapid and safe assessment platform to evaluate the neutralizing efficacy of therapeutic antibodies against emerging SARS-CoV-2 variants using a pseudovirus-based system. ○ Development of Pseudovirus-Based Neutralization Evaluation System To evaluate the neutralizing efficacy of therapeutic antibodies against emerging SARS-CoV-2 variants, we developed a pseudovirus-based neutralization assay. Production conditions were optimized to ensure high-yield generation of pseudoviruses. Viral titer and infectivity were systematically assessed to confirm assay reliability. Performance validation was conducted by comparing our pseudovirus-based assay platform to a widely utilized plaque reduction neutralization test (PRNT), affirming its applicability. ○ Comparison of High-Throughput Screening (HTS) and PRNT Methods for Neutralization Assays HTS (105.9 ng/ml) and PRNT (95.22 ng/ml) yielded nearly identical IC50 values, indicating HTS’s suitability for large-scale neutralization screening. This system allows for rapid and efficient evaluation of antibody candidates against novel variants. ○ Securing Commercially Available COVID-19 Therapeutic Antibodies Commercial antibodies were obtained by inserting antibody gene sequences into expression vectors and subsequently overexpressing them in animal cells. The neutralization potency of these antibodies against variant strains was assessed, confirming their effectiveness relative to previously reported results. These therapeutic antibodies served as controls in the validation of the neutralization evaluation platform. Enhancement of Neutralization Potency Against Emerging Variants Through Novel Antibody Development ◈ Objective: To develop a novel antibody platform targeting diverse epitopes to address the diminished efficacy of existing monoclonal antibodies against novel variants. ○ Efficacy Evaluation of Antibodies Discovered Using Phage Display Eighteen antibodies were screened using phage display technology, and three promising candidates (#4, #11, #38) were selected for further evaluation. These antibodies demonstrated high neutralization potency against the Delta variant in cell-based assays but were less effective in animal models. ○ Development of IgG Antibody Cocktail Therapy The efficacy of two different antibodies (IgG-D, IgG-I) targeting distinct epitopes was evaluated individually and in combination. In cell-based assays, the combination therapy exhibited excellent neutralizing activity against BA.5, XBB.1.5, and BA.2 variants. The therapeutic efficacy of the antibodies was validated in a mouse model, with the combination providing broader protection than the monoclonal antibodies alone. SPR and AI structural analysis predicted noncompetitive binding to different epitopes. ○ Discovery of Inhaled Antibodies by Switching (IgG to IgA) Intravenous IgG antibodies were converted to inhaled IgA forms (D.A2, I.A1, and D.dA2), and the neutralization efficiencies of these generated antibodies were compared. IgA antibodies exhibited enhanced binding affinity to the spike protein and demonstrated comparable or superior neutralization activity compared to IgG antibodies. In animal studies, IgA antibodies showed improved therapeutic efficacy when administered in combination, highlighting the potential of isotype switching in therapeutic development. ○ Development and Evaluation of Bispecific Antibody (IgG scFv, Tandem scFv-Fc) Bispecific antibodies (IgG scFv, Tandem scFv-Fc), composed of variable regions of two different antibodies (IgG-D, IgG-I), were developed to target both wild-type and mutant strains. Bispecific antibodies exhibited higher neutralizing efficacy than IgG antibodies alone or in combination. Notably, the Tandem scFv-Fc construct showed the highest therapeutic efficacy in both cell-based and animal model evaluations. These results suggest that bispecific antibody platforms offer a promising strategy for countering evolving viral variants. Conclusion and Future Directions - The pseudovirus-based neutralization assay platform is a valuable tool for the rapid and safe evaluation of antibody therapeutics against SARS-CoV-2 variants. - This study highlights the effectiveness of antibody cocktails, isotype switching, and bispecific antibody therapies in countering SARS-CoV-2 variant resistance. - Contributing to the development of next-generation therapeutic antibodies, it lays the foundation for rapid responses to emerging infectious diseases, enhancing national health security.
