A co-culture model of liver organoids and macrophages was developed to study HCV infection and metabolic-associated steatohepatitis (MASH). By co-culturing macrophages with liver organoids, it was confirmed that the macrophages acquired characteristics similar to Kupffer cells, which are liver-specific macrophages. Additionally, bioinformatics tools were used to analyze that hepatic stellate cells remained in an inactive state, closely resembling those in human liver tissue. Our co-culture model is applicable not only for studying normal liver physiology but also for investigating pathological conditions. It effectively mimics the progression of infections and metabolic diseases, demonstrating its utility in evaluating drug mechanisms and predicting clinical outcomes. These findings were published in a scientific journal. By utilizing advanced human lung organoids, a dual infection model for coronavirus and influenza virus was established and validated, demonstrating the high utility of this platform for studying various human respiratory viral diseases. It was confirmed that human lung organoids derived from pluripotent stem cells exhibit a high degree of human similarity, indicating their significant potential for future lung disease research. Through this research the expected outcomes are followings, first, Advanced organoids are not only useful for infectious disease modeling but also hold great promise for the development of diverse disease models. By enhancing organoid fabrication techniques, human diseases can be more accurately replicated. Second, Through infection modeling, a novel research platform was established to study the interactions among tissue cells, immune cells, and pathogenic viruses. This platform is expected to have extensive applications in infectious disease research. Finally, By evaluating the human resemblance of organoids and analyzing their characteristics, criteria for the standardization of organoid quality assessment can be proposed for future research.
