The ultimate goal of this research project is to establish the foundation for identifying therapeutic targets for kidney diseases by developing differentiation techniques for kidney organoids derived from induced pluripotent stem cells (iPSCs) and creating models that simulate kidney diseases. The research was conducted over two years. In the first year, the focus was on generating kidney organoids derived from human iPSCs and developing kidney disease models using these organoids. In the second year, the aim was to replicate the established kidney organoid protocols and provide training in organoid generation techniques to the commissioning department. At the request of the commissioning department, three national stem cell lines from the National Stem Cell Regeneration Center were obtained. Using a total of six different pluripotent stem cell lines—comprising one embryonic stem cell line and five induced pluripotent stem cell lines—a differentiation method for culturing kidney organoids was established, and the functional characteristics of the organoids as kidney models were thoroughly analyzed in vitro. Moreover, to address the limitations of existing kidney organoids—such as difficulties in long-term culture and frozen-thawing after differentiation—a method was developed for expandable kidney organoid-derived tubule organoids. These tubule organoids, which exhibit human-like responsiveness, were exposed to high concentrations of glucose and inflammatory cytokines for up to 30 days, revealing cellular changes in the kidney. The epithelial-mesenchymal transition (EMT) response of tubule epithelial cells, a key phenomenon in kidney failure, was also observed. The results of this research and the organoid production techniques were shared with the commissioning department through online and offline meetings, and a standardized protocol was also submitted in document form. This study successfully established reproducible kidney organoid production techniques and demonstrated the potential for kidney failure modeling using these organoids. The outcomes provide a foundation for various kidney disease research applications, including the identification of disease-specific target factors, drug efficacy and toxicity evaluations, and elucidation of pathophysiological mechanisms.
